中文摘要: 病毒感染触发的细胞凋亡既是机体抗病毒防御机制之一,也是病毒逃避免疫应答和/或诱发持续感染一种策略。研究发现,以ATCC VR-2332株为代表的美洲型猪繁殖与呼吸综合征病毒(PRRSV)和以LV株为代表的欧洲型PRRSV均可在体内外诱导细胞凋亡,且诱导机制与病毒致病机制有关。PRRSV是通过与细胞膜表面上的特异性受体结合,利用细胞内吞作用而感染易感细胞,非肌肉肌球蛋白II型重链A(NMHC-IIA)是新确定的PRRSV的辅助受体。本研究利用人工微小RNA(artificial microRNAs, amiRNAs)干扰Marc-145细胞 NMHC-IIA的表达,通过探讨PRRSV感染对敲低NMHC-IIA的细胞凋亡信号传导的影响及其机制,进而阐明NMHC-IIA在PRRSV感染过程中的生物学功能,为进一步揭示PRRSV的致病机理提供材料,为猪繁殖与呼吸综合征的防控提供理论依据。

英文摘要: Cell apoptosis caused by virus infection is not only an antiviral defense mechanism, but also a strategy for virus escaping immune response and/or developing persistent infection. Research found that American type porcine productive and respiratory syndrome virus(PRRSV), represented by strain ATCC VR-2332, and Europe type PRRSV, represented by strain LV, all could induce cell apoptosis both in vitro and in vivo, and the induction mechanism was related with virus pathogenesis. PRRSV infected susceptible cells by binding with specific receptor on the surface of the cell membrane and cells endocytosis. Nonmuscle myosin heavy chain II-A(NMHC-IIA) is a PRRSV receptor identified recently. In this study, NMHC-IIA expression of Marc145 cells was interfered using artificial microRNA, then the effect and its mechanism of PRRSV infected NMHC-IIA knockdown cells on cell apoptosis signaling transduction will be explored. Then, biological function of NMHC-IIA in PRRSV infection will be clarified and PRRSV pathogenesis mechanism will be further revealed by analyzing those results.
