中文摘要: 金黄色葡萄球菌(金葡菌)是重要的人类致病菌,可产生多种致病因子对宿主产生破坏作用从而引起多种病症。金葡菌致病因子的分泌受到其自身调控网络的精细调控, 包括一系列的转录因子(Sar蛋白家族为主)、二元信号系统、群体感应系统以及非编码小RNA等,但其详细机制的阐述仍存在不足。我们的前期工作发现金葡菌gdpS基因的缺失能够影响一系列致病因子的表达,在金葡菌致病性调控中作用关键。进一步研究发现,gdpS基因行使对protein A的调控功能,并不依赖于其编码的蛋白形式,而是依赖于其转录出的RNA形式,即gdpS编码出的mRNA不仅可以作为信使RNA当作蛋白翻译的模版,它还是一个具有调控功能的RNA形式,类似于我们所知的具有调控功能的非编码RNA。我们希望阐明gdpS调控protein A及其他下游靶点的详细分子机理,从而深化和丰富对金葡菌致病性调控的认识。

英文摘要: Staphylococcus aureus is an important human pathogen, which can cause many infectious diseases by producing a range of virulence factors. The production of these factors is modulated by a complicated regulatory network, including a series of transcription factors (mainly including the Sar homologue), two-component regulatory systems, quorum sensing systems and small non-coding RNAs. However, the research of the detailed regulatory mechanisms still remains insufficient. Our previous work found that the inactivation of gdpS affected the production of many virulence factors, suggesting that gdpS plays an important role in virulence regulation in S. aureus. Furthermore, we found that gdpS can regulate the production of protein A, not by the protein coding by gdpS, but by the RNA it transcribes. It indicates that the mRNA coding by gdpS not only acts as the messenger RNA, but also can work as a regulatory RNA, like the known regulatory non-coding RNAs. In this study, we will investigate the detailed molecular mechanism of how gdpS regulates protein A and other downstream targets, in order to adding the knowledge of virulence regulation in S. aureus.
