中文摘要: 在抗病毒转基因研究中,以往曾尝试利用多种来源于病原的抗性基因如外壳蛋白基因、运动蛋白基因、复制酶基因等,然而由于抗性水平低、抗谱窄、抗性逐步丧失等原因,这些基因都没有获得更广泛的应用。近年研究表明,利用RNA沉默机制获得的抗病毒水稻植株抗性水平可达免疫,具有较大应用潜力。然而此单一抗性策略的应用会面临病毒变异导致水稻抗性丧失的风险。针对抗病毒策略极度匮乏的现状,有必要构建新型抗病毒策略。本项目拟通过病毒与水稻的互作研究来解析水稻体内参与病毒侵染的相关途径,并通过多种分子生物学手段分析这些调控途径中处于关键位置的基因或miRNA,在证实这些基因或miRNA在病毒侵染、增殖、扩散过程中的潜在功能、明确它们是有助于病毒侵染还是抑制病毒侵染的基础上,通过增强水稻体内的抑制病毒侵染的途径或弱化水稻体内有助于病毒侵染的途径来增强水稻对病毒抗性,构建新型抗病毒策略。

英文摘要: Pathogen-derived resistant genes, such as coat protein, movement protein, or replicase gene of viruses, had been used into producing the resistant plant agaist viruses. However, these strategies were not applied widely because of the limit resistance. Recent decadal research has shown that RNA silencing-based resistant plants have the high level, even immune resistance to viruses, indicating the applicable potential. Meanwhile, the wide use of the single strategy has to face the risk that the resistance might lose for the viral variation. Hence, new strategies are needed. In this proposal, we will uncover the pathways of plant related to Rice stripe virus infection, basing on the studies on the interaction between virus and rice, and then, identify the key components (proteins, genes or miRNAs) in these pathways and understand their functions either helpful or inhibitory for viral infecion. Subsequently, we explore the new strategies to obtain the high resistant plant through strengthening the inhibitory function or weakening the helpful function for viral infection. We hope develop several new strategies that are applicable into molecular breeding of resistant plants.
