中文摘要: 巨噬细胞在天然免疫防御和获得性免疫应答中起着重要作用。巨噬细胞是许多多糖的主要作用靶细胞,多糖通过其特异性寡糖片段与巨噬细胞表面的受体结合后,活化启动细胞内信号转导途径从而产生效应功能。对于受体的研究可以阐明多糖类药物的作用原理和生物信号转导机制,为多糖类药物设计、靶点的选择和用药方案的确定提供理论依据。申请人项目组前期研究发现,仙茅多糖对巨噬细胞激活作用确切。但目前仙茅多糖一级结构研究不深入,高级结构未见涉及,构效关系及对巨噬细胞激活作用机制不明确,具有进一步深入研究价值。本项目旨在对仙茅活性多糖纯级分进行酶法降解,酶解片段进行巨噬细胞活性筛选。探讨仙茅多糖纯级分及其酶解片段一级结构、高级结构与活性的关系;探寻仙茅多糖中与巨噬细胞受体结合的特异性活性寡糖片段,并较为系统地探讨活性片段激活巨噬细胞的作用机制。本项目的顺利实施将为多糖类药物的开发及构效关系的研究奠定较好的理论基础。

英文摘要: Macrophages play an important role in the natural immune defense and adaptive immune response. Macrophages are the primary target cells many polysaccharides . Polysaccharides usually bind with receptors on the surface of the macrophages through specific segments of oligosaccharides, and then activate intracellular signal transduction pathways to start function and produce effects. The function principle and biological signal transduction mechanism of polysaccharides could be clarified through the studies on their receptors. Which could also provide a theoretical basis for the polysaccharide drug design, choice of targets and the determination of the regimen. The preliminary study on his project showed that curculigo polysaccharide had the function of macrophage activation. Curculigo polysaccharide can be further research because its primary, senior structure,structure-activity relationship and mechanism of macrophage activation are still not clear. This project aims to study the enzymatic degradation of the pure active curculigo polysaccharide, then the screening macrophage activity of the hydrolysate. The relationships between the activities and the primary, senior structures will be studied. The specific active oligosaccharide fragments of curculigo polysaccharide which binded with macrophage receptor and their mechanism of macrophage activation will also be systematically explored. The implementation of the project might provide significant theoretical basis for the polysaccharides drug development and their structure-activity relationship studies.
