中文摘要: 我国是茶叶生产大国,拥有丰富的茶树花资源。多糖具有抗衰老、免疫调节与抗肿瘤等多样的生理功能,但目前国内外对茶树花多糖的研究报道较少,且缺乏系统性。基于我们良好的前期研究基础,本项目拟研究茶树花多糖的免疫与抗肿瘤活性及其结构与活性的关系,具有重要的理论意义。主要研究内容如下:(1)建立茶树花活性多糖提取、分离与纯化的方法;(2)通过细胞培养等技术筛选具有免疫活性和抗肿瘤活性的茶树花活性多糖,并在此基础上进行相应的免疫与抗肿瘤的动物体内实验研究,明确茶树花活性多糖的免疫调节与抗肿瘤活性;(3)采用UV、Smith降解等方法对茶树花活性多糖进行结构表征;(4)利用纳米粒度仪等分析多糖溶液的特性粘度、粒度分布、比旋光度等茶树花多糖的溶液行为,利用原子力显微镜、动态光散射仪等分析茶树花多糖的分子链构象,以阐明茶树花多糖的活性与结构的关系。

英文摘要: China produces a plenty of tea each year, accordingly, there is a rich resource of tea flower. Polysaccharides have been demonstrated to exhibit a variety of physiological functions, such as anti-aging, immune regulation and anti-tumor activity. However, only a few, but not systemic studies on polysaccharides from tea flower (TFPS) are available. Based on our previous studies, the objectives of this project are to investigate the immunostimulatory and anti-tumor activities of TFPS and stuy the relationship between its structure and bioactivity. The main contents of this project are as following: (1) To develop efficient procedures for the extraction, separation and purification of TFPS; (2) to screen the fractions of TFPS that have the immunostimulatory and anti-tumor activities by in vitro assays, further, to test their bioactivities by using corresponding in vivo animal models, in order to get the immunostimulatory and anti-tumor activities of TFPS clear; (3) To characterize the structure of TFPS by using methods of UV, Smith degradation and etc.; (4) To analyze the solution behaviors of TFPS, such as intrinsic viscosity, particle size distribution, optical rotation and etc. by using nano-particle size analytical instrument; to investigate the molecular chain conformation of TFPS by using atomic force microsco
