中文摘要: 绵羊肺炎支原体是引起绵羊及山羊非典型肺炎的重要病原,其分布广泛,危害严重。但是,至今对其重要毒力因子及免疫原知之甚少。粘附素是各种致病性支原体的重要毒力因子和免疫原,已成为阐明支原体致病机制、开发疫苗及建立血清学诊断技术的重要研究对象。然而,至今对绵羊肺炎支原体的粘附素尚无任何研究。本项目在前期对绵羊肺炎支原体进行全基因测序及生物信息学分析的基础上,选择预测得到的粘附素P113蛋白作为研究对象,开展两方面的试验研究:一是分析其在绵羊肺炎支原体粘附宿主细胞中的作用,确定其是否为粘附素;二是评价其所诱导的体液及细胞免疫应答水平,分析其免疫原特性,并通过免疫和攻毒保护试验分析其免疫保护作用。本研究旨在利用试验研究,证明P113蛋白作为粘附素及免疫原科学推论,为阐明绵羊肺炎支原体的致病机理提供理论依据,为开发有效的亚单位苗和建立有效的诊断工具提供基础。

英文摘要: Mycoplasma ovipneumoniae is etiological angent of atypical pneumoniae in sheep and goats. However, little is known about improtant immnuogens and virulence factors of this pathogen. Adhesin is one of the most important virulence factors and immunogens of all pathogenic mycoplasmas. Therefore, adhesin has became the most concerned protein when researching on pathogensis, vaccine development and establishment of serological diagnostic methods. However, nothing is known about adhesin of mycoplasma ovipneumoniae,so far. Previously, we sequenced the whole genome of mycoplasma ovipneumoniae and deduced that protein P113 is a potential adhesin.In this project,a serial methods will be applied to verify two scientific hypothesis: (1) P113 protein is a major adhesin and play an important role in adherence to host cell; (2) P113 is a predominant immunogen and is able to induce effective humoral and cellular immune response that can protect hosts against mycoplasma ovipneumoniae infection. The purpose of this project is to research the characterization of P113 protein as an adhesin and immunogen, so as to provide basis for clarifing pathogenisis, developing vaccines and useful diagnostic methods.
