中文摘要: 前期我们发现,SIRT1能够通过抑制肿瘤细胞EMT进程,进而抑制肿瘤的转移(Journal of the national cancer institute,2013; Journal of cell science,2013);进一步的microRNA表达谱芯片结果发现,SIRT1能够可能参与调控miR-15b-5p的转录;而同时miR-15b-5p下游靶向重要的EMT相关蛋白(如E-cadherin、RECK等)。在此基础上我们提出假设: SIRT1可能通过调控miR-15b-5p的转录进而参与结直肠癌转移过程。本研究以前期建立的裸鼠结直肠癌转移模型为基础,结合结直肠癌相关细胞系及结直肠癌临床组织标本,从整体动物模型、细胞模型及临床标本等方面阐明SIRT1通过miRNA调控结直肠癌转移的分子机制。

英文摘要: We previously found that SIRT1 could repress cancer metastasis by down-regulating cancer cell EMT(Journal of the national cancer institute,2013; Journal of cell science,2013); Recently, the microRNA-microarray results showed that SIRT1 may participate in regulating the transcriptional activity of miR-15b-5p; Moreover, miR-15b-5p downstream targets some important proteins involved in EMT(such as:E-cadherin、RECK).Therefore, we hypothesized that SIRT1 could regulate transcriptional activity of miR-15b-5p and thus play an important role in colorectal cancer metastasis. We will investigate the role of SIRT1 and miR-15b-5p in pathogenesis and progression of CRC in the model of colorectal cancer metastasis in nude mice, clinical samples of CRC and cell models.
