中文摘要: 生殖支原体(M.genitalium, Mg)是最小的原核细胞型微生物,寄生在泌尿生殖道,常引起泌尿生殖道感染,如各种高危性的性传播疾病等。核酸适配体是人工合成的核酸序列,能以高亲合力同相应靶物质(如Mg)结合。基于磁致伸缩原理设计的磁致弹性传感检测方法,是一种优秀的非接触式无线传感检测技术,特别适用于透过一定障碍对物质进行实时、原位检测。本项目拟通过SELEX技术筛选出Mg的核酸适配体,在磁致弹性无线传感片上修饰Mg核酸适配体,特异性识别并与Mg结合,将Mg的量转化为磁致弹性传感片的表面载荷质量变化,从而使磁致弹性传感片共振频率发生变化。通过在一定条件下Mg的量与共振频率变化之间的定量关系,研制出可透过实物障碍对Mg进行特异检测的磁致弹性无线传感装置,并建立相应的检测方法学。该法可很好地保护受检者隐私。通过本项目研究,还可为其他病原体核酸适配体的筛选和无线传感检测研究提供理论和实验依据。

英文摘要: Mycoplasma genitalium (Mg) is the smallest prokaryotic cell microorganisms and parasitizes in the urogenital tract. It is a common cause of urinary and reproductive tract infections, such as high-risk of sexually transmitted diseases. The aptamer is a synthetic nucleic acid sequence which have the high affinity with the corresponding target materials (such as Mg). Based on the magnetostrictive designed, magnetoelastic sensor detection is an excellent non-contact wireless sensing technique, especially suitable for real-time, in situ detection of substances through certain obstacles. The project intends to select Mg aptamer by the SELEX technology, modified wireless magnetoelastic sensor with Mg aptamer, specific identification and combined with Mg, Mg amount of payload mass on the magnetoelastic sensor chip surface changes, and the resonant frequency of magnetoelastic sensor chip change. Under certain condition, the quantitative relationship between changes in volume and resonance is easy to set up. The wireless magnetoelastic sensing devices would be developed for specific detection of Mg through the physical barriers. The appropriate testing methodology would be established,too.
