中文摘要: 微阵列已成为在基因组水平研究基因表达的最有力工具。但是现有实验存在下列特点:一是多集中于模式生物(例如小鼠、大鼠)和人,而在猪等其它动物方面却因芯片技术平台尚未完善、成本昂贵等而没有广泛开展;二是几乎都存在着样本数太小的问题,但在模式生物和人方面对于相同问题经常存在许多重复或者类似研究。而猪的繁殖性能意义重大,但是现有的QTL定位和候选基因分析进展缓慢。有鉴于此,本研究针对GEO和Arrayexpress数据库中已有模式生物和人等的繁殖相关微阵列数据的元分析进行了以下工作:①#23376;宫内膜异位症芯片数据的通路富集分析及整合;②#21754;乳动物(小鼠,人和牛)胚胎早期发育过程相关芯片的整合分析;③#22522;于小鼠基因表达谱的比较分析识别乳腺发育过程关键通路的研究。另外,基于芯片元分析结果,我们针对HIF-α23478;族和VEGFA基因多态性与猪繁殖性状进行了相关研究。这些都有助于我们更深入地研究猪繁殖性状的遗传机理。

英文摘要: Microarray technology has become one of the most powerful tools for studying gene expression in genome wide. However, there are the following limitations in the current microarray experiments. Firstly, most of them are focused on the model animals (such as mice and rats) or humans, but less were on pigs or other dometical animals since poor platforms and high cost. Secondly, the sample sizes of all the experiments are so small, wheras there are frequently many repeat or similar studies on the same problem of the model animals or humans.On the other hand, porcine reproduction traits are so important, but the progress on QTL mapping and candidate gene approach is slow. Therefore, we have done the following works based on the meta-analysis on microarray datasets of animal models or human from the databases of GEO and Arrayexpress:①A cross-study gene set enrichment analysis identifies critical pathways in endometriosis; ②Meta-analysis on microarray data of early embryonic development of three mammalian species (mouse, human and bovine);③Comparative gene expression analysis in mouse models for identifying critical pathways in mammary gland development. Moreover,based on the results of our meta-analysis, we then performed the further study on the relationship between gene polymorphisms of HIF-αamily and VEGFA with porcine reproduction traits.All of these may play important roles in further researches in animal functional genomics related to porcine reproduction.
