中文摘要: 以盐酸普萘洛尔、布洛芬等药物的手性拆分为应用背景,本课题拟研究生物大分子DNA自组装结构手性特征的调节规律,针对不同药物对映异构体,设计构建具有不同识别位点和立体选择性的核酸手性模板;研究不同对映异构体在手性模板诱导下的成核生长机理,优化降温速率、溶剂组成、晶种类型等结晶条件,以实现基于DNA手性识别的对映异构体的择优结晶拆分。本课题首次基于核酸组装结构的手性识别特征,研究开发用于药物对映体拆分的新途径,研究成果可以促进医药结晶和手性拆分技术的发展;而且核酸手性自组装规律与药物对映体的手性识别机理对于分子生物学的研究也具有重要意义。

英文摘要: Aiming to develop a chiral resolution route of drugs including propranolol, ibuprofen, etc., the regulation mechanism of chirality will be studied for biomacromolecular DNA self-assembly, in order to construct chiral DNA templates with specific recognition sites and stereoselectivities towards different drug enantiomers. The nucleation and growth mechanism of different enantiomers will be studied under the orientation of chiral DNA templates. Optimizing the crystallization conditions involving the cooling rate, the solvent, the seed species, etc., the preferential crystallization of drug enantiomers will be studied through the chiral recognition of DNA templates. It is the first time to develop a novel chiral resolution route for drug enantiomers under the orientation of chiral recognition of DNA templates, the results will promote the development of medicine crystallization and chiral separation techniques. Moreover, studies on the chirality of DNA self-assembly and the recognition mechanism towards drug enantiomers are also fundamental to the investigations on molecular biology.
