中文摘要: 前体RNA剪接是基因转录产物去除内含子,连接外显子的过程。RNA剪接是真核生物基因表达的重要调控方式,是生物体响应发育及各种胁迫的重要机制。我们在前期的工作中克隆了一个编码含有RNA结合域RRM的基因TDR1(Tolerant to Drought1),并得到了该基因缺失突变体tdr1-1。tdr1-1明显增强植物对干旱的耐受,但是对ABA敏感性明显提高;在突变体中过量表达TDR1全长cDNA能够恢复所有上述表型,表明该突变体的上述表型确实是由TDR1功能缺失造成的。本研究将在此研究基础上,通过生理生化、分子生物学及遗传学等方法,对TDR1的表达模式、亚细胞定位及RNA结合活性进行分析,并对其参与调控ABA信号的机制进行深入研究,同时通过大通量RNA-seq分析寻找可能的TDR1的剪接底物,通过转基因方法确定该底物在ABA信号通路中的功能,最终将解析植物体在转录后水调节ABA信号的作用机。

英文摘要: Pre-mRNA splicing is a modification of an RNA after transcription, in which introns present in primary are removed and exons are joined. It is a fundamental mechanism of constitutive eukaryotic gene expression and an important regulation of gene expression. We have cloned a gene named TDR1, which encodes a RRM-containing protein in Arabidopsis, and obtained a loss-of-function mutant tdr1-1. Because tdr1-1 showed increased tolerance to drought, the corresponding gene was called TDR1: Tolerant to Drought). However, the mutant was hypersensitive to ABA. In this work, we will analyze the expression pattern, subcellular localization and RNA binding activity of TDR1. Our research will focus on the TDR1 functions in the regulation of ABA signal transduction using a combinatorial approach of plant physiology, molecular biology and genetics. Through the transcriptional analysis, we will search for the substrates of TDR1, and validate the function of the substrates in ABA signaling pathway by a genetic approach. The final goal is to uncover the mechanism that regulates ABA response at the post-transcriptional level in Arabidopsis.
