中文摘要: 面对北冰洋严酷的环境压力,放线菌必须拥有相当独特的生理、生化特性才能在其中生存。因此,这些放线菌可能已进化出与温带、热带放线菌不同的次生代谢产物合成途径,从而产生结构类型新颖、生物活性显著的次生代谢产物,但目前对其知之甚少。本课题将综合运用微生物学、分子生物学和药物化学的方法,分析北冰洋太平洋扇区不同海洋沉积环境中放线菌多样性及其产新结构化合物的潜力,旨在获取北冰洋太平洋扇区沉积环境放线菌资源,揭示其物种多样性和生物医药应用潜力;同时结合海洋沉积环境基础理化性质分析,研究不同沉积环境对放线菌的丰度、多样性及其次生代谢产物合成能力的潜在影响。这些工作,将为大规模获取北冰洋国际公共海域海洋放线菌资源和进一步开发利用北冰洋放线菌资源奠定理论基础。

英文摘要: Arctic marine ecosystems have developed in a rather cold and severe climate, suggesting that the selective pressure on microorganisms comprising parts of such systems must have been quite unique. Because of this, it seems likely that these microorganisms have developed secondary metabolites biosynthesis pathways that differ from those utilized by temperate and/or tropical microorganisms. However, the information about Arctic marine actinobacteria is rare. In this proposal, the methods of microbiology, molecular biology and medicinal chemistry will be comprehensively used to analyze the actinobacterial diversity and its potential for production of the new structural compounds in different marine surface sediments from the Pacific Arctic Ocean. The following subjects will be involved in this proposal: i) The abundance of the actinobacteria in sediments from Pacifc Arctic Ocean; ii) The diversity of the actinobacteria in sediments from Pacifc Arctic Ocean; iii) The secondary metabolite biosynthesis capacity of the actinobacteria in sediments from Pacifc Arctic Ocean; iv) The relationship between sedimentary environmental factors and the diversity of actinomycetes and their secondary metabolite synthesis capacity.These efforts will help increase our understanding on the actinobacteria form Arctic Ocean, as well as
