中文摘要: 丘脑背内侧核(MD)是重要的内脏和躯体活动的整合中枢,前额叶皮层(PFC)是哺乳动物最高级别的联合皮层。我们前期的工作发现:束缚-浸水应激(RWIS)大鼠MD、PFC内Fos阳性表达显著增强;损毁PFC后RWIS大鼠胃运动受到显著抑制;向PFC内注射HRP并给予RWIS,48h后在MD内发现HRP标记神经元。由此我们提出:MD-PFC神经环路可能参与RWIS调控。本项目以Wistar雄性大鼠为材料,用免疫组化、Western-blot、束路追踪、电生理学等技术从神经元类型、突触可塑性、纤维投射及神经元放电等方面进行研究,重点解决以下问题:1)MD内参与RWIS反应的神经元类型,即××能神经元或××敏感神经元;2)RWIS时MD内神经元的活动规律。研究结果将阐明MD在RWIS中的作用,为进一步揭示应激致胃机能紊乱的中枢神经机制提供实验依据。

英文摘要: As we all known, the mediodorsal nucleus of thalamus (MD) is the integration center of the visceral and physical activities while the mammalian prefrontal cortex (PFC) is the most-superior cortex. Our previous researches demonstrated that there was remarkable Fos expression in the PFC and MD of rats induced by restraint water-immersion stress (RWIS), and gastric motility was inhibited after bilateral PFC lesions during RWIS. Furthermore, when we injected horse radish (HRP) into PFC and gave rats a RWIS, 48 h later neurons labeled by HRP were found in the MD. So, we hypothesized that the MD-PFC circuits may be involved in the response to RWIS. If so, 1) What is the phenotype nature of activated neurons in the MD during RWIS? 2) What is the regularities of activated neurons in the MD during RWIS ? To determine the above questions, immunohistochemical, western-blot, tract-tracing, and electrophysiological methods would be used in the present research. The achievements in the research would determine the role of the MD and the results would provide an important foundation to probe into the central mechanism of gastric dysfunction induced by RWIS.
