中文摘要: 细胞衰老是器官和个体衰老的基础,也是抑制肿瘤发生的机制。SIRT6具有去乙酰化酶和单ADP-核糖基化酶活性,能够对包括衰老在内的多种生命活动进行调控。PTEN/ p27Kip1途径是调控细胞衰老的一个经典通路。本课题拟深入研究SIRT6延缓细胞衰老的分子机制即SIRT6对p27Kip1蛋白质稳定性的影响及其翻译后修饰(泛素化和乙酰化)之间的反馈调节关系。阐明这一问题不仅有助于揭示细胞衰老的分子机制,还可为延缓衰老及临床肿瘤的诊断和预防提供实验依据。

英文摘要: The sirtuin SIRT6 is a NAD-dependent histone deacetylase that is implicated in gene regulation and lifespan control. But whether SIRT6 has the function to influence cell viability and senescence under non-stressed conditions in human diploid fibroblasts is far from unknown. Our data showed that enforced SIRT6 expression promoted cell proliferation and antagonized cellular senescence. Furthermore, we demonstrate that SIRT6 antagonized endogenous p27Kip1 expression through improving ubiquitin-proteasome-mediated degradation. Finally, we suggest that SIRT6 markedly rescues the replicative senescence induced by p27Kip1 via potentiating p27Kip1 degradation. Therefore, our study provides a new molecular mechanism for the regulation of cellular senescence.
