中文摘要: 细胞存活/死亡机制的失衡是导致肿瘤细胞耐药的根本,寻找调控细胞存活/死亡的关键分子开关在克服肿瘤耐药机制研究中具有重要意义。我们的研究发现多功能衔接蛋白p62高表达与肿瘤顺铂耐药性正相关,为进一步深入探讨p62在顺铂耐药中的作用,在综述文献和前期工作的基础上,本研究的着眼点放在p62调控重要存活/死亡信号关键分子的泛素化上。受体相互作用蛋白1(RIP1)可与p62直接结合,泛素化形式决定其发挥存活还是死亡功能。我们假设p62和RIP1的相互作用通过影响NFκB活性参与肿瘤顺铂耐药机制。本研究以顺铂作用下人卵巢癌SKOV3细胞(亲代)和SKOV3/DDP细胞(顺铂耐药)为实验模型,分别构建p62 si-RNA、p62 ZZ结构域突变、UBA结构域突变表达载体,观察其对RIP1泛素化形式、IKKs-NFκB活性信号通路活性影响,从调控RIP1泛素化角度探讨p62在顺铂耐药机制中的作用。

英文摘要: The imbalance between cell survival and cell death is a relatively important fundamental factor resulting in tumor drug resistance,thus looking for a master switch that regulates cell survival and death is of great significance to overcome this puzzle.Our study found that high expression of the multifunctional adapter protein p62 positively correlates with resistance of tumor cells to cisplatin;to further explore the role of p62 in cisplatin resistance,based on literature readings and the preliminary work,a key focus of this study is to figure out how p62 regulates the ubiquitination of critical molecules involving in the death-survival signal.The ubiquitination level of receptor interacting protein 1 (RIP1),which could directly bind to p62,determines its role as a pro-death or pro-survival factor.We assume that p62 interacting with RIP1 may participate in tumor resistance to cisplatin by affecting NFκB activity.In this study,the parental SKOV3 and cisplatin-resistant SKOV3/DDP human ovarian carcinoma cell lines are adopted,through inhibiting the expression of p62 by small interfering (si)RNA or by constructing expression vectors containing a zinc finger (ZZ) domain or the UBA domain mutations within p62,we aim to observe how p62 regulates the ubiquitination of RIP1 and the activity of NFκB,thereby clarifying th
