中文摘要: 蛋白磷酸酶2A(PP2A)是脑内调节tau蛋白去磷酸化的主要蛋白磷酸酶,PP2A活性降低可能是导致AD脑中tau过度磷酸化、神经元丢失和死亡的主要原因。山茱萸环烯醚萜苷(CIG)是我们从山茱萸中提取的主要有效部位,前期研究首次发现,CIG能够明显拮抗tau蛋白过度磷酸化,提高PP2A活性,但是其确切的作用机制尚不清楚。本项目的科学假说是:CIG可能通过调节PP2Ac翻译后修饰,促进脑内PP2A激活,抑制tau蛋白过度磷酸化。主要研究内容为:(1)应用GSK-3β转染细胞模型,结合siRNA技术,进行CIG对PP2Ac磷酸化和甲基化修饰调节的机制研究及药物靶标研究。(2)应用MAPT*P301L转基因致tau过度磷酸化小鼠模型,结合PP2Ac条件敲除小鼠,进行CIG在体内对PP2Ac翻译后修饰的影响及拮抗tau蛋白过度磷酸化的机制研究。本项目将为AD的药物研究提供新的策略和潜在治疗靶点。

英文摘要: A down-regulation of protein phosphatase 2A (PP2A), the major tau phosphatase in human brain, contributes to tau hyperphosphorylation and neurodegeneration in Alzheimer's disease (AD). Abnormal post-translational modification of PP2A catalytic C subunit (PP2Ac) is reported to reduce PP2A activity. Previous studies in our laboratory indicated that Cornel iridoid glycoside (CIG), the main component extracted from Cornus of?cinalis, attenuates tau hyperphosphorylation by increasing PP2A activity. However, the pharmacological mechanism of CIG remains unclear. The hypothesis of our project is that CIG may activate PP2A activity to attenuate tau hyper-phosphorylation by regulating post-translational modification of PP2Ac. The main contents in our study includes (1) to investigate the drug target and mechanism of CIG on regulating phosphorylation and methylation of PP2Ac by GSK-3β transfected N2a neuroblastoma cells and siRNA technology; (2) to investigate the in vivo effect of CIG on post-translational modification of PP2Ac and the mechanism of CIG on tau dephosphorylation by using MAPT*P301L transgenic mice and PP2Ac conditional knock-out mice. This project may provide a new strategy and potential effective targets for AD drug therapy.
