中文摘要: 前庭传出神经可以通过乙酰胆碱(Ach)和降钙素基因相关肽(CGRP)等递质调节外周前庭传入神经的兴奋性,而前庭传入神经元也可以直接调节前庭传出神经元的功能。但是目前并不了解前庭核传入、传出神经元之间的直接投射通路是依靠何种递质起调节作用以及如何调节该通路的功能。既往研究提示Ach和CGRP是前庭中枢系统传入、传出神经元的主要递质,它们与前庭代偿密切相关。本课题拟完成:1.应用膜片钳电生理技术明确Ach、CGRP是否为该通路的神经递质及其作用。2.应用免疫组化和免疫荧光法,电刺激兴奋和毁损抑制前庭核传入神经元后,分别观察前庭传出神经元Ach和CGRP的变化。通过上述研究,明确Ach和CGRP是否是前庭传入、传出神经元直接投射通路的主要神经递质;力图阐明前庭核传入神经元的兴奋性发生改变后,如何通过Ach和CGRP来调节前庭传出神经元的兴奋性,从而进一步探讨前庭代偿的可能发生机制。

英文摘要: Vestibular efferent neurons could regulate the excitability of peripheral vestibular afferent nerve by acetylcholine (Ach) and calcitonin gene-related peptide (CGRP), as well as other neurotransmitters. Vestibular afferent neurons could aiso directly regulate the function of the vestibular efferent neurons. But now people do not understand what kinds of transmitters are being in the direct projection from vestibular afferent neurons to vestibular efferent neurons and how these transmitters adjust the regulating function of this pathway.Previous studies suggested that Ach and CGRP are the major neurotransmitters of afferent and efferent neurons in central vestibular system, They are closely related to vestibular compensation.The project intends to complete: 1. To identify whether Ach and CGRP are the neurotransmitters in above pathway and what are their roles by patch-clamp electrophysiology. 2. By the experimental methods of immunohistochemistry and immunofluorescence: ⑴Excited afferent neurons through electrical stimulation in the vestibular nuclei, observed the changes of Ach and CGRP in vestibular efferent neurons. ⑵Damaged the vestibular afferent neurons, further observed the changes of Ach and CGRP in vestibular efferent neurons.Through this research, we try our best to clarify how Ach and CGRP regulate the
