中文摘要: 猪苓是一种珍贵的药用真菌,其菌核入药,猪苓菌核在我国用药已达两千多年的历史。然而,随着猪苓菌核药用范围和用量的扩大,加上野生猪苓菌核资源的商业采挖以及缺乏有效的保护,使得猪苓菌核的生境遭受严重破坏,野生的猪苓菌核资源濒临枯竭。 本项目研究内容包括:通过成功构建的猪苓菌丝形成菌核抑制差减杂交cDNA文库,对表达序列标签进行数据分析;利用实时荧光定量PCR技术分析与氧化应激、信号转导途径等相关基因响应猪苓菌丝形成菌核过程中的基因表达分析;利用RACE技术克隆全长基因,并对猪苓菌核形成相关基因进行初步的功能验证。 上述研究工作的开展,不仅为揭示猪苓菌生长发育规律的基因表达特征奠定基础,同时为解析猪苓菌核形成及对药用真菌猪苓菌核的人工栽培工作提供理论依据。此项工作不仅具有重要的理论意义,而且具有潜在的应用价值。

英文摘要: Polyporus umbellatus is one of the most widely used and precious medicinal fungi, which has been used to treat many diseases such as edema and acute nephritis for more than two thousand years. However, wild sclerotia of P. umbellatus are almost in danger of extinction due to unrestricted commercial harvesting, ineffective protection, and severe habitat destruction. This study is conducted to analyze expressed sequence tags (ESTs) data based on suppressive subtractive hybridization cDNA library of P. umbellatus sclerotia and hyphae which has been successfully constructed. Then gene expression of oxidative stress and signal pathways which are probably associated with P. umbellatus sclerotia forming from hyphae will be detected by real-time quantitative fluorescence polymerase chain reaction (PCR). Full-length genes will be cloned by means of RACE. At last, Fuctional verification of genes associated with P. umbellatus sclerotial formation will be conducted. This study will provide us for gene expression characteristics in uncovering growth and development in P. umbellatus. On the other hand, it will help us to understand mechanisms of sclerotial formation and serve as a basis for implementing mass production of P. umbellatus.
