中文摘要: 河南省是我国冷冻食品产业第一大省,金黄色葡萄球菌在冷冻食品中屡有检出,严重影响人们的食用安全。本项目组前期研究结果发现,冷冻胁迫下金黄色葡萄球菌细胞受到明显的亚致死损伤,但仍能够存活相当长时间,并推测与相关基因的表达调控密切相关。本项目将以未受冷冻胁迫细胞为对照,采用全基因组表达谱基因芯片结合Realtime PCR技术、二维聚丙烯酰胺凝胶电泳(2-D PAGE)结合MALDI串联飞行时间质谱(MALDI TOF/TOF)技术,重点研究:①冷冻胁迫下亚致死金黄色葡萄球菌转录组差异表达谱;②冷冻胁迫下亚致死金黄色葡萄球菌蛋白质组差异表达谱,从而发掘冷冻胁迫耐受相关的基因。本项目研究结果不仅能够丰富食源性致病菌亚致死和胁迫耐受理论,为从相关基因表达调控的角度揭示金黄色葡萄球菌冷冻胁迫耐受机制奠定基础,也能够为冷冻食品中金黄色葡萄球菌控制技术的建立提供理论依据。

英文摘要: Henan province is the leading province of frozen food industry in China, whereas, the most commonly detected Staphylococcus aureus causes serious food safety issues for frozen food. According to our previous studies, many S. aureus cells could keep alive for a long period although having suffered sublethal injury, which was presumed to be closely related with the expression of certain genes. In present research project, as compared with non-stressed cells, the whole genome expression profile gene chip combined with realtime PCR and 2 D polyacrylamide gel electrophoresis (2-D PAGE) combined with matrix-assisted laser desorption/ionization time-of-flight (MALDI TOF/TOF) assays will be employed to take following research, respectively: 1, the differentially transcriptional expression characterization of sublethal S. aureus under frozen stress; 2, the differentially proteome expression characterization of sublethal S. aureus under frozen stress, which is aimed to explore the frozen resistance related genes. The research results will not only be helpful to enrich the theories of sublethal effect and stress resistance of foodborne pathogens, reveal the molecular mechanism of S. aureus during resistance to frozen stress from the perspective of related genes expression, but also offer a theoretical basis for the establi
