中文摘要: 细粒棘球蚴病(包虫病)是一种重要的世界性人畜共患寄生虫病,严重危害我国畜牧业以及人类健康。细粒棘球绦虫的中绦期幼虫(细粒棘球蚴)存在可育和不育两种包囊,只有可育囊才能完成其生活史。细胞凋亡可能会导致不育囊这一特殊生物学现象的形成,但参与的基因及调控机制尚不明确。本研究拟对绵羊体内细粒棘球蚴可育囊/不育囊进行转录组测序与基因差异表达分析,挖掘与细胞凋亡相关的基因;并从中筛选2-3个候选基因进行克隆与表达,通过对体外培养的细粒棘球蚴原头蚴进行靶基因的RNA干扰,研究其功能及在不育囊形成中的作用,并探讨细粒棘球蚴不育囊形成的分子调控机制,为人和动物细粒棘球蚴病新型疫苗的开发及防控策略提供基础资料和新的思路。

英文摘要: Cystic echinococcosis (CE) is an important parasitic zoonosis worldwide, which seriously influence the production of livestock husbandry and the health of human. Echinococcus granulosus have fertile and infertile cysts, but only the fertile cysts can finish their life cycles. Apoptosis may cause the phenomenon, but its molecular mechanism of this special biological phenomenon that E. granulosus develop infertile cysts in its intermediate hosts is unclear. In our study, we analyze different gene expression (transcriptome) of fertile/infertile cysts of E. granulosus from sheep and digging out the apoptosis-related genes based on the differentially expressed genes and gene function enrichment analysis. Then we clone and express 2-3 candidate genes among them. To investigate the function and mechanism of these genes in the formation of infertile cysts, protoscolexs were cultured in vitro with specific siRNA targeting these recombinant proteins. Thus, we can make insights intothe mechanisms of the infertile cysts formation, and provide basic data and newstrategies for the vaccine development, prevention and controlling of CE.
