中文摘要: Beta-arrestins是G蛋白偶联受体信号通路中的重要信号分子。近年研究发现,beta-arrestins除了作为经典的G蛋白偶联受体信号转导过程中的抑制因子外,还可以作为信号分子直接的参入细胞的信号转导过程,调控多种神经递质的生理效应。提示beta-arrestins可能参与多种脑高级功能的调控。本研究的目的是探讨beta-arrestins在突触可塑性和学习记忆中的作用。本课题采用beta-arrestins基因敲除品系小鼠,在多种学习记忆模型中检测beta-arrestins对小鼠学习记忆能力的影响,同时采用全细胞膜片钳记录技术研究相应脑区突触可塑性的改变,并探讨相关机制。研究结果揭示了beta-arrestin在调节神经突触可塑性和恐惧记忆形成中的重要作用并阐述了其机制,论文发表在Proc Natl Acad Sci USA上,并被Nature China选为研究亮点特别介绍。

英文摘要: Beta-arrestins are important signal molecules in the G protein coupled receptor signal pathway. The most great research development on the beta-arrestins over the past few years is their multifaceted roles as signaling adaptors and scaffolds to take part in cellular signal transduction directly and regulate many kinds of physiological effects including neurotransmitter induced signal. All these imply that beta-arrestins are involved in many brain function modulation. The present research will explore the role of beta-arrestins in the synaptic plasticity and learning and memory. We will investigate the ability of learning and memory between beta-arrestins knock out and wild type mice in a few of behavior models. Meanwhile, we will aslo study the synaptic plasticity related with beta-arrestins knock out mice phetotype and find out the mechanism. The article published in Proc Natl Acad Sci USA, has been evaluated highly by Nature China.
