中文摘要: 由铁盐催化的有机反应近年来已经受到了有机化学家们的高度重视,这是因为相对传统过渡金属催化剂而言,铁盐的成本更低,对环境更加友好,而且保存更为方便。但是总体来说,铁催化的反应依然还有很大的发展空间。本项目拟对铁盐/碘代异丙烷体系催化的有机镁试剂参与的串联反应展开研究,对适合该催化体系的底物进行筛选,并且对反应条件进行全面的优化,合成该串联反应的产物(如二氢茚类、吲哚啉类和苯并呋喃类衍生物);结合质谱分析,通过对比实验及动力学实验,进一步对该类型反应进行机理探索;最后,将该方法应用于具有潜在药学活性分子的合成中,并对这些分子进行初步的药学和生物学活性筛选。本课题对于扩展铁催化的反应领域,发展新的有机合成方法,具有重要的理论意义,同时,对化学工业界也有重要的参考价值。

英文摘要: Given its low price, non-toxicity and environmentally benign character, which often offers notable advantages in terms of sustainable chemistry, iron catalysis represents hence an attractive alternative to other commonly used transition metals. However, judging from the explored research, the iron-catalyzed organic reactions still have a long way to go. In this proposal, we are planning to make a detailed study on the iron-catalyzed tandem reaction (radical cyclization then cross-coupling), which concerns on the the preparation of the precursors of indane derivatives,indoline derivatives and benzofuran derivatives, and the reaction condition optimization for the synthesis of the target molecules. The study on the Mass analysis, control experiments and the kinetic research of the reaction speed, will tell us some evidence of the possible mechanism. Furthermore, the proposed protocol will be applied to the synthesis of some potentially pharmaceutically and biologically interesting molecules, the synthesized compounds will be subsequently submitted for pharmaceutical and biologycal analysis.
