中文摘要: 脑源性神经营养因子(BDNF)是神经营养素家族成员之一,其主要作用是促进神经系统发育。近年来研究表明,BDNF及其受体TrkB在雄性动物睾丸中也有分布,然而,BDNF在睾丸发育中的作用,国内外尚无报道。我们的研究发现,BDNF可以调节牛精子的生理功能;BDNF分泌紊乱可能与男性不育有一定关系。在预实验中发现,小鼠睾丸间质细胞能分泌BDNF,并且BDNF可以调节间质细胞合成睾酮,同时能促进类固醇急性调节蛋白(StAR)的表达。因此,提出"BDNF通过促进StAR蛋白表达调节睾丸间质细胞分泌睾酮进而影响睾丸发育"的假设。为阐明此假设,本课题拟采用Western blot、Real-time PCR等技术,从整体-细胞-分子三个层面,探索BDNF对睾丸间质细胞合成睾酮的影响及机制。本课题将为认识睾酮合成的机理提供理论依据,期望为睾酮分泌紊乱引起的雄性不育提供潜在的预防和治疗靶点。

英文摘要: Brain-derived neurotrophic factor (BDNF) is a member of neurotrophins, which plays important roles in the development of nervous system. Recent evidences showed that BDNF and its receptor TrkB localized in the Leydig cells of testis. But, there is not any evidence about the role of BDNF in testis. Our laboratory showed that BDNF could regulate physiological functions of bovine sperm, and abnormal expression of BNDF in the testis could be relative with male infertility. The preliminary results showed that leydig cells in the testis of mice could secret BDNF, and BDNF could increase testosterone production. Meanwhile, BDNF could increase the expression of steroidogenic acute regulatory protein (StAR) which plays an important role in testosterone production. According to our results, we hypothesize that BDNF can regulate testosterone production through increasing the StAR expression, and further affect testicular development. To verify the hypothesis, techniques of Western blot, Real-time PCR, etc. will be used to study the role and signal transduction of BDNF in testosterone production from different aspects. The project will provide new theoretical evidences for the mechanism of testosterone production, and further provide potentially therapeutic targets for male infertility caused by abnormal production of testo
