中文摘要: 新型锌指蛋白ZBTB20是BTB/POZ家族的新成员,在神经、消化等多个系统具有重要的生物学功能,但在心脏中的作用尚未见报道。在本课题组建立的ZBTB20全身基因敲除小鼠模型中,我们观察到心脏发育和心功能出现异常。为进一步了解ZBTB20在心脏中的生物学功能,我们利用Cre/loxp技术建立了心脏组织特异性ZBTB20敲除(HZB20KO)小鼠模型,发现该模型小鼠心脏结构未见明显异常,但是心肌收缩功能却明显增加,在单个心肌细胞中也观察到收缩幅度和钙瞬变幅度大于对照组小鼠心肌细胞。这些前期结果表明ZBTB20在心肌收缩中具有重要的调控作用。本课题拟在此基础上,结合分子生物学、电生理学等多种研究手段,深入分析HZB20KO小鼠心肌细胞中与收缩相关的通道和分子表达和功能的改变,阐明ZBTB20调控心肌收缩的分子和电生理机制,可望形成原创性科研成果,进一步完善人们对ZBTB20生物学功能的认识。

英文摘要: ZBTB20, a new member of the BTB/POZ zinc finger family, plays a variety of important roles in multiple systems such as nervous and digestive systems, but its function on heart has not been established. In ZBTB20 global knockout mice, we observed cardiac malformations and cardiac function was impaired. To address the function of ZBTB20 in the heart, we generated heart-specific ZBTB20 knockout (HZB20KO) mice by Cre/loxp system. HZB20KO resulted in increased cardiac contract function, without affecting heart structure. In isolated cardiomyocytes, the amplitude of contraction and calcium transients of HZB20KO mice was bigger than control mice. These results indicate that ZBTB20 play an important role in the regulation of myocardial contractility. In the present study, we further analysis the change of expression and function of channels and molecules related with myocardial contractility by techniques of molecular biology, electrophysiology, and et al, which may clarify the mechanisms of the regulation of ZBTB20 on myocardial contractility, form primary achievements in scientific research, and improve the recognition on the biological function of ZBTB20.
