中文摘要: 作为乙型病毒性肝炎开放读码框之一的HBx基因与宿主细胞转录激活、细胞周期调控等许多重要生命活动密切相关。本申请建立在申请人前期对HBx转基因小鼠定量蛋白组学研究发现HBx引发肝细胞恶性转化与激活CDC42信号通路关系密切的基础之上,设计采用在基因敲除或过表达CDC42基因、SILAC正反标记细胞结合蛋白组学技术,同时采用靶向蛋白组学与定量蛋白组学结合的方法研究CDC42基因功能,旨在从蛋白质相互作用和基因网络调控的角度更准确地揭示CDC42信号激活在HBx介导肝细胞恶性转化进程中的作用机制,同时也将加深对HBV作用机制的理解。

英文摘要: HBx gene, one ORF of hepatitis b virus, was closely related to multiple important biological function of host cells, including the transcriptional activation andcell cycle regulation. We found that HBx induced the malignant transformation of hepatocytes through the activation of CDC42 signaling pathway by the large scale quantitative proteomics study with SILAC labeled mice and theHBx transgenic mice. However, the mechanism of the activation of CDC42 signaling pathway during the occurance and development of liver disease induced by HBx remains study. We propose to apply quantitative proteomics study to reveal the CDC42 signal from the regulation of protein interaction and gene network in HBx inducedmalignant transformation process under the gene knockout or overexpression of CDC42 in HBx transfected cells. The study will also enhance the understanding of the mechanism of action of HBV.
