中文摘要: 手足口病(HFMD)是由肠道病毒71型(EV71)感染导致在婴儿与儿童中常见的疾病,包括:发烧、腹泻、皮肤红疹或水泡。EV71感染还可引起无菌脑膜炎、脑干脑炎和脊髓灰质炎样麻痹等多种神经系统相关疾病,严重者会导致死亡。但目前人们对EV71感染导致神经系统损伤的机制还不清楚。本项目结合前期工作基础,拟通过两个方面的研究来解释EV71对神经细胞功能的调控作用。一方面研究病毒直接感染神经细胞后通过调控神经细胞多巴胺及胞内炎症因子的表达从而影响神经细胞的功能。另一方面探讨病毒是否通过激活体内的免疫细胞产生特定炎症因子并通过血脑屏障作用于脑内的神经细胞,从而影响神经细胞的功能。通过本项目研究,确定EV71感染对神经细胞功能的影响,并初步阐明EV71感染导致患者神经系统损伤的分子机制。

英文摘要: Hand, foot and mouth disease (HFMD) caused by enterovirus type 71 (EV71) infection is common in infants and children's, including fever, diarrhea, skin rash or blisters. EV71 infection can also lead to diseases associated with aseptic meningitis, brain stem encephalitis, and poliomyelitis like paralysis and other nervous system, severe cases can lead to death. But the mechanisms underlines EV71 infection leading to nervous system injury is unclear. This project combines the foundation, proposed by the two aspects of research to explain the role of EV71 in regulation of nerve cell function. First, we will investigate the effects of EV71 infection on the expression of inflammatory cytokines and intracellular dopamine, and thus on neuronal function of neural cells. Second, we will determine whether the virus affects the function of nerve cells through the activation of the immune cells to produce specific inflammatory factors, which overcome the blood brain barrier to enter the brain nerve cells. Thus, through this project, we will determine the effect of EV71 infection on neural cell function, and elucidate the molecular mechanisms by which EV71 infection in patients leading to nervous system injury.
