中文摘要: 轮状病毒(rotaviruses,RVs)是引起婴幼儿及动物病毒性胃肠炎的重要病原体。跨物种传播是轮状病毒毒株多样性的重要原因,给轮状病毒疫苗的研发和使用带来巨大挑战。最新的研究发现组织血型抗原(histo-blood group antigens,HBGAs)有可能作为RVs的结合受体在其跨物种传播中扮演重要作用。P[19]型RV可以感染猪和人,具有跨种传播的能力,但具体机制未明。本项目旨在研究和分析猪源和人源P[19]型RV VP8*蛋白与HBGAs的结合特性及其结构基础的异同,探讨型P[19]轮状病毒跨物种传播的分子机制,为阐明RV致病机理及RV疫苗的研制提供基础和依据。

英文摘要: Rotaviruses (RVs) are one of the most important pathogens for the viral gastroenteritis in human and animals. RV cross-species transmission contributes to the RV strain diversity, which brings great challenge for the development and use of RV vaccine. It is reported that histo-blood group antigens (HBGAs) may be the receptors and/or attachment factors for the RVs recently and then play a vital role in the RV cross-species transmission. P[19] RVs have been detected both in human and pigs and show distinct gene assortments and cross-species transmission. However, the mechanism remain unclear. In this study, we will focus on the characterization of the HBGAs binding specificity of both porcine and human origin P[19] VP8* proteins with saliva- and oligosaccharide-based assays. Furthermore, the crystal structures of VP8* and HBGAs will be investigated to reveal the structural basis of the binding patterns, which is significant to clarify the mechanism of cross-species transmission and lay the foundation for the RV vaccine design.
