中文摘要: 生物材料与血液和组织的不相容性常常限制了它们的应用。尽管一些材料的表面改性方法已被广泛研究以降低材料介入体内所引起的不相容性,但在实际应用中取得的进展却依然有限。本研究的目标是利用化学反应将脂质体完整、稳定的固定于固体材料表面,而生物分子通过与脂质体表面特定官能团的特异反应被修饰到材料表面,利用生物分子的生物活性提高生物材料与血液以及组织的相容性,达到降低免疫系统应激反应和抑制血栓形成的作用。这一目标是通过两个平行操作完成,脂质体完整、稳定的固定和生物膜模拟的功能化。其特点是首先,由脂质体覆盖的材料表面模拟了生物膜表面磷指非特异吸附的特性改善了材料与血液的相容性;其次所引入的生物分子血栓调节蛋白和肝磷脂模拟了黏多糖和膜蛋白抑制血栓形成的特性。这些特点为生物医学表面材料的研究和设计提供了新的途径。

英文摘要: Current biomaterials suffer from the well-known problem of surface induced incompatibility and thrombosis and thus have limited applications. The proposed study focus on a new bio-membrane mimetic interface nanomaterial designed to enhance blood-compatibility and to prevent thrombus formation on biomaterials. This proposal is based on a central hypothesis that immobilized intact liposomes with spatially controlled placement of heparin and thrombomodulin will minimize non-specific adhesions and inhibit thrombosis formation. The proposed research will be focused through two operational paradigms, 'immobilized intact liposome' and 'bio-membrane mimetic antithrombogenesis'. These operational paradigms will provide guidance in cytomimetic designs and focus specific research on: (1) bio-membrane mimetic interface nanomaterial that mimic the non-adhesive property and anticoagulant function of phospholipids on bio-membrane surfaces to improve blood compatibility; and (2) bio-membrane mimetic interface nanomaterial that mimic bio-membrane antithrombotic glycosaminoglycans and proteins to inhibit thrombosis formation. The proposed immobilized liposome-based bio-membrane mimetic interface nanomaterial will be assembled by bio-orthogonal and chemical selective lipsome immobilization/stabilization/surface functionalization strategy.
