中文摘要: 基于不断发展的测序技术,最近几年我们可以用前所未有的深度和广度来研究基因组功能。越来越多的研究证实真核生物中存在大量非编码RNA(包括长链非编码RNA、假基因和环状RNA),它们在细胞和组织分化、染色质修饰、转录调控、免疫系统、表观修饰和疾病等方面起着非常重要的作用。然而目前只有少数非编码RNA的功能进行了实验验证,而绝大部分非编码RNA只是在转录组数据中被鉴定出来。为了探索非编码RNA是否具有功能,我们选择了15个代表性动物和一个外群物种酵母来研究非编码RNA,将从表达水平、剪切机制、序列保守性、结构和基因组特征等方面对非编RNA进行系统的分析,阐述非编码RNA的保守性和进化并揭示它们的功能。这项研究将为我们提供非编码RNA起源和进化方面的线索。此外,我们将通过基因敲除或过表达的方法在细胞水平对个别高度保守的非编码RNA进行功能验证。

英文摘要: Based on the advanced sequencing technologies, we can study genome functions in unprecedented depth and breadth in recent years. More and more studies have proved the abundance of noncoding RNA (including long non-coding RNA, pseudogene and circular RNA) in eukaryotes, which play important roles in differentiation of cell and tissue, chromatin modification, transcription regulation, immune system, epigenetic modification and disease. However, only a few of noncoding RNA's functions have been experimentally verified, whereas the majority are just identified by transcriptome data. To explore whether these noncoding RNA function or not, we selected 15 representative animals and yeast as the out-group to study noncoding RNA and illustrate their conservation, evolution and function from expression, RNA splicing, sequence conservation, structure and genome feature. This study will provide some clues for the origin and evolution of noncoding RNA. In addition, we will do functional experiments for some highly conserved noncoding RNA using gene knockout or over expressing method in cell level.
