中文摘要: 对生命遗传信息的解读是现代生物学研究关注的重点之一。核酸结构对基因表达的调控信息是遗传信息的重要组成部分。G四链体是一类在基因组中丰度和广度都非常高的的核酸结构,且在基因表达的多个阶段,尤其是在蛋白翻译过程中具有重要调控功能。目前对G四链体的研究多为模拟生理环境和细胞中的实验,对体内G四链体的形成机制和功能仍远未阐明。本项目计划利用果蝇模型,结合基因组分析数据和细胞培养等体外模型,系统研究:1)蛋白翻译过程中G四链体的形成及检测;2)G四链体对蛋白翻译的调控;3)G四链体结合小分子及相互作用蛋白对相关过程的影响。此项研究不但可以丰富G-四链体领域的研究,进一步揭示G-四链体的生物学功能,还能为解析其他核酸结构对基因表达的调控提供借鉴和新思路,为解读生命遗传信息提供新的线索。

英文摘要: Interpretation of genetic information of organism is one of the focal points of modern biological research. And the regulatory role of secondary structures of nucleic acids play in gene regulation is essential part of genetic information. G-quadruplex is a species of secondary structure widely present in genome with large amount, which is crucial in regulation of gene expression, especially translation. Previous studies were mostly carried out on simulated physiological or intracellular environment. The mechanism of G-quadruplex formation and function of G-quadruplex in vivo are not clear. We plan to establish a system on basis of genome-wide analysis and experiments in cell and Drosophila to study: 1) the formation and 2) the regulatory role of G-quadruplex during translation, and 3) the effect of G-quadruplex binding molecules and G-quadruplex interaction protein. This study will broaden the knowledge of G-quadruplex research and further reveal the biological function of G-quadruplex. Moreover, it will provide new ideas and clues for analysis other secondary structure of nucleic acids.
