中文摘要: 针对严重危害我国人口健康素质的大骨节病软骨损伤及其细胞信号转导障碍的关键问题,基于近年来所获大骨节病软骨细胞差异蛋白鉴定的研究进展,采用体外软骨细胞立体培养、共聚焦显微镜、Western Blot等分子生物学技术,集中研究大骨节病软骨细胞信号转导和细胞行为模式中起重要作用的细胞骨架分子的变化,确定软骨细胞内细胞骨架分子的定位及其与细胞死亡和凋亡的关系,观察细胞骨架分子和氧化应激分子的基因和蛋白质表达,进而确定大骨节病软骨细胞骨架重组RhoA和Rac1途径的活化状态。藉此为准,比对细胞、蛋白和基因不同层次细胞骨架重组的变化,通过观察T-2毒素对正常软骨细胞骨架的影响和补硒干预细胞骨架重组途径的作用,明确大骨节病软骨细胞骨架重组的主要途径及环境因素的作用,为阐明软骨细胞损伤过程中细胞骨架重组的机制及其可调控措施,提高预防和治疗大骨节病技术水平,提供科学依据。

英文摘要: The cartilage damage and signal transducton disorder in chondrocyte are key issuses of Kashin-Beck disease (KBD) which is serious harmful endemic disease to the health in china. Based on the research progress of KBD in recent years with received differential expressed proteins in KBD chondrocyte, we use the molecular biology technique with culture the chondrocyte with 3D model, confocal microscopy, western blot and so on to study the molecular changes of the cytoskeleton that plays an important role in cell signal transduction and cell behavior patterns, to definite the cytoskeleton molecular location in cell and its effection on cell death and apoptosis. To study the gene expression and protein expression of cytoskeleton and oxidative stress related molecular to definite the RhoA/Rac1 pathway activity in KBD chondrocyte. The main cytoskeleton remodeling pathway of KBD and the environment factor effection will be demonstrated by those comparative studys at different levels as cell, protein and gene, and compare with T-2 toxin treated normal chondrocyte and selenium treated KBD. These results will provide scientific basis to clarify the mechanism of the cytoskeleton remodeling in the process of cartilage cell damage and its control measures to improve the prevention and treatment of Kashin-Beck disease technical
