中文摘要: 细胞生物力学性质研究是细胞生物学研究中的热点内容,对生物学基础研究以及临床医学应用均有重要意义。目前细胞生物力学性质测量技术中存在自动化程度低、细胞生物力学性质表征形式单一等问题。本课题以细胞骨架生物力学性质测量为目标,拟建立一种基于视觉传感的微纳力和位移测量技术,实现在较大载荷下细胞力学性质的高精度测量;同时,采用双微悬臂法,实时测量探针夹持器与样品表面之间的漂移并对其进行补偿,解决测量设备中普遍存在的机械漂移问题。此外,为实现自动测量,开发基于计算机图像处理的细胞显微图像分割技术,实现细胞轮廓的提取和定位。在以上工作基础上,建立一种新型的细胞生物力学性质自动测量系统并以人体乳腺癌细胞MCF 10A、MCF7、和MDA-MB-231为研究对象,探索融合细胞形态学的细胞生物力学性质的多参数表征方法。

英文摘要: The study of cell mechanics is important for both fundamental research in cell biology and cell based clinical applications and is gaining more and more attentions. Currently the experiments for cell mechanics measurement are mostly conducted manually and the statistic analysis of the cell mechanics merely includes other cell properties. The purpose of the project is to develop a technique to measurement the cell mechanics for cell cytoskeletons. To solve the above mentioned issues, a novel technique is proposed to measure force and displacement in micro/nanoscale with assistance of the vision sensing technique. With the proposed method, the large load can be preciously applied to the target cells, which allows for the measurement of cell mechanics for the entire cytoskeletons. Moreover, a two beam method will be developed to measure and then balance the mechanical drift commonly existed in the measurement system in real time. Additionally, new algorithm will be developed to achieve accurate cell segmentation in cell optical images for the purpose of cell boundary detection and localization. With the above designs, a one-of-the-kind instrument equipped with novel algorithms and software will be developed to perform automatic measurement of cell mechanics.
