中文摘要: 通过对原发性肝细胞癌样本全基因表达谱芯片的检测分析,以及对肝癌肿瘤细胞系的检测,我们发现基因真核生物翻译起始因子3H(EIF3H)在肝癌样本中高表达。本项目拟在肝癌细胞上通过RNA干扰技术,研究内源性EIF3H对肿瘤增殖,周期,凋亡等影响,初步确认EIF3H的基因功能,通过免疫组化在大样本上,确认EIF3H的表达和肝癌肿瘤临床及病理的相关性。同时计划通过深入的动物实验,多细胞的功能验证,以及EIF3H过表达后的功能研究,使得其功能更加明确。在机制方面,拟通过特定下游基因的蛋白合成速度和微弱mRNA的翻译效率的检测,明确被其调控的下游基因,特别是肿瘤功能相关基因的表达变化以及EIF3H的调控机制。希望通过本项目,对寻找肝癌诊断及预后判断指标和干预靶点奠定基础。

英文摘要: Hepatocellular carcinoma (HCC) is the common malignant tumor and cause of cancer-related death, it is important to understand the molecular mechanisms that contribute to tumor initiation or progression of liver cancer. Through whole gene expression microarray detection of HCC samples and cell lines, we found eukaryotic translation initiation factor 3H (EIF3H) was highly expressed in HCC. In our project here, first we will knockdown the expression of EIF3H through siRNA to study whether endogenesis EIF3H play important roles on liver cancer cells proliferation, cell cycle and apoptosis; then we will confirm the relation between expression of EIF3H and clinical pathology in liver cancer. At the same time, we will further study the function of EIF3H using animal models and various cell function assays. To investigate the mechanism, we will define the downstream target genes of EIF3H through special protein synthesis detection and mRNA transcription detection, especially we will focus on how EIF3H regulate the cancer-relatived gene. We hope this study could lay a foundation for HCC diagnosis, prognosis, and therapy.
