中文摘要: 人体多种疾病,例如癌症、动脉硬化、糖尿病、肝炎、白内障、心血管病、老年痴呆、关节炎等,都与体内自由基过量密切相关,通常需要从多方面摄取足够量的抗氧化剂来消除多余的自由基,以保持身体健康。因此,抗氧化剂的开发一直是保健品、化妆品等行业最主要的研发方向之一,是市场最重要的功能性诉求之一。根据前期研究表明:稀土和嘧啶酮甲磺酸均有抗氧化活性,稀土与有机配体结合形成配合物后可以在很大程度上改变、修饰和增强稀土的生物活性。因此,本项目研究嘧啶酮甲磺酸稀土配合物的合成与抗氧化活性。通过检测它们的还原能力,清除羟基、DPPH和ABTS等自由基的能力,螯合Fe2+能力;检测它们与BSA的相互作用、对蛋白质损伤的保护作用、对肝微粒体脂质过氧化的抑制作用以及对肿瘤细胞生长的抑制作用来衡量受试化合物的生物活性;建立可靠的CoMFA和CoMSIA模型来指导目标化合物的分子设计与结构修饰。

英文摘要: Many diseases, such as cancer, arteriosclerosis, diabetes, hepatitis, cataract, cardiovascular diseases, Alzheimer's disease, arthritis, etc., have correlation with the excess of radicals inside body. In order to keep healthy, people need to ingest sufficient antioxidant from various sources. The development of antioxidant, therefore, has been one of the major research areas and market demands in health and cosmetic industry. The preliminary research shows that both rare earth metals and pyrimidinonyl methanesulfonic acids have the property of antioxidation, which can be changed, tuned or improved to a large extend by the coordination of rare earth metals and organic ligands. The purpose of the project is to investigate the synthesis and antioxidation of complexes of pyrimidinonyl methanesulfonic acids and rare earth metals. In order to study the biological properties of these compounds, we will look into their reduction, ability of cleaning hydroxyl, DPPH and ABTS, chelation of Fe2+. In addition, these complexes' interaction with BSA, protection of proteins, effects against rat liver microsomallipid peroxidation and inhibition of cancer cells will be studied. CoMFA and CoMSIA models will be constructed to guide the molecular design and structural modifications of target compounds.
