中文摘要: 妇科虚寒证是临床常见的病证之一,项目申请人前期研究发现能量代谢障碍、神经-内分泌-免疫网络紊乱和氧化应激损伤构成了妇科虚寒证的主要病理学基础,其中氧化应激损伤是妇科虚寒证较为特异的指标之一。抗氧化应激体系Nrf2信号和促凋亡的NF-κB信号在氧化应激损伤中发挥了正-反两方面的调控作用。因此本研究通过构建大鼠虚寒证模型及寒冷刺激细胞模型,靶向于卵巢颗粒细胞,探讨:1)氧化应激损伤与妇科虚寒证卵巢颗粒细胞凋亡和内分泌功能紊乱之间的关联;2)金匮温经汤能够通过上调Nrf2信号/抑制NF-κB信号,拮抗凋亡和促进内分泌调节平衡,发挥抗氧化应激效应以治疗妇科虚寒证。这一研究为解析妇科虚寒证证候学本质,丰富“异病同治”的理论提供了新的思路和实验依据。

英文摘要: Gynecological asthenia-cold syndrome is the more prevalent disease in clinical. In the previous study, We demostrated that energy metabolism disorder, NEI abnormality and oxidative stress are composed to main pathological basis of Gynecological asthenia-cold syndrome. And oxidative stress could be a specific marker of Gynecological asthenia-cold syndrome. Nrf2/HO-1 signal is known to orchestrate antioxidant and detioxidant response to NF-κB signal in oxidative stress. Therefore, this study proposes to use asthenia-cold rat model combined with cell oxidative stress model induced by cold, to explore 1) the relationship of apoptosis and endocrine disorder in ovarian granulosa cell with oxidative stress; 2) the anti-oxidative mechanism of the Jingui Wenjing Decoction in anti-apotosis and promoting endocrine homeostasis by regulating Nrf2 and NF-κB signal. This study provides a new way and some valuable information for exploring the Syndrome resolution of “Gynecological asthenia-cold syndrome” and the principle of “Different disease with the same treatment” in Jingui Wenjing Decoction.
