中文摘要: 染色质重塑是通过表观遗传修饰调控基因表达的重要方式。染色质重构蛋白依靠水解ATP 释放的能量改变染色质结构,是染色质重塑的主要调控者之一。拟南芥中有42个sucrose non-fermenting two(SNF2)家族的染色质重构蛋白,它们中大部分蛋白的功能未被研究。本项目前期通过对拟南芥自体免疫(autoimmunity)突变体bon1-1表型抑制突变体的遗传筛选,发现染色质重构蛋白CHR5与抗病免疫反应相关。基于此我们拟通过:1)遗传筛选CHR5参与拟南芥抗病免疫反应调控的互作基因;2)转录组高通量测序(RNA-Seq)和染色质免疫共沉淀(ChIP)鉴定CHR5参与抗病免疫反应调控的靶标基因;3)酵母双杂交和双分子荧光互补(BiFC)鉴定CHR5参与抗病免疫反应调控的互作蛋白等研究,进一步解析CHR5在拟南芥抗病免疫反应中的功能,为其它植物染色质重构蛋白功能研究提供理论依据。

英文摘要: Chromatin remodeling is one important epigenetic modification that regulates gene expression by changing the structure of the chromatin. Chromatin remodeling ATPase protein, a major player in the process, utilizes energy derived from ATP hydrolysis to alter histone-DNA contacts within the nucleosome. There are 42 putative SNF2 ATPases in Arabidopsis, many of which have un-known functions. Recently, chromatin remodeling ATPase protein 5 (CHR5) was identified as a regulator of the plant immunity by the genetic suppressor screening of the autoimmunity mutant bon1-1. This study aims at further elucidating the regulatory mechanism of CHR5 in Arabidopsis immunity. Specific objectives are: 1) isolation of genetic interactor of CHR5 in Arabidopsis immunity by suppressor screen; 2) identification of direct regulatory targets of CHR5 by ChIP (chromatin immuno-precipitation) in Arabidopsis immunity; 3) identification of CHR5 interaction proteins by yeast two hybrid screen and bimolecular fluorescence complementation. The above study will elucidate the regulatory mechanisms of CHR5 in Arabidopsis immunity and provide a theoretical basis for the function study of other plant chromatin remodeling ATPase proteins.
