中文摘要: 鳜鱼因其味道鲜美,肉质细嫩,营养价值高,已成为水产养殖业中重要养殖名贵鱼类之一。肌卫星细胞是肌肉生长和修复过程中新肌纤维的来源,但关于肌卫星细胞的维持静息状态和激活分化的分子机理还未阐明。miRNAs在肌肉发育分化过程中扮演着极其重要的调控功能。本课题拟从miRNAs在鳜静息态和激活态肌卫星细胞中的差异表达分析入手,筛选和证实对鳜肌卫星细胞静息状态维持起重要作用的特异性miRNAs及其靶基因,建立特异性miRNAs上游转录调控因子、特异性miRNAs及其靶基因的调控网络,揭示特异性miRNA信号通路对维持卫星细胞静息状态的作用机理。其研究结果不仅有助于我们更深一步阐明鱼类肌肉胚后生长发育调控机制,而且将为我国鱼类高效健康养殖提供重要理论依据。

英文摘要: As one of the most commercially important fish in China, the Chinese perch exhibits a quite lot of advantages, such as good meat texture, full scale of amino acid profile, especially high content of essential and flavor amino acids. Muscle satellite cells provide with new cellular resources for muscle growth and self-renewal. Increasing evidences have suggested that miRNAs are the key regulators of the myogenesis.In this project, we will use the Chinese perch as model system to reveal the molecular mechaisms of maintenance of the muscle satellite cell quiescence by microRNAs. The project will be specifically focused on the following aims: 1)to analyze and identify the significantly expressed miRNAs between the quiescent and activated muscle satellite cells using miRNA microarray; 2) to investigate the function of the specific miRNAs and their target genes;3)to construct the regulatory networks between the specific miRNAs and their transcription factor and target genes.Collectively,these studies will make significant contribution to the current understanding of muscle satellite cell activation and quiescence. More importantly, these findings also provide the technical implications for aquacultural application.
