中文摘要: 精神分裂症阴性症状病理机制不明,缺乏有效的治疗措施,部分患者出现严重的脑功能残疾。最近的研究认为脑内小胶质细胞(microglia, MI)过度激活产生的神经炎症可能是精神分裂症阴性症状的病因之一。核因子κB(NF-κB)信号通路、丝裂原激活蛋白激酶(MAPK)信号通路、蛋白酪氨酸激酶/信号转导子与激活子(JAK-STAT)信号通路被认为是MI内与神经炎症密切相关的信号通路。我们的前期研究成功建立了“发育期MI异常激活的精神分裂症大鼠模型”。本研究拟在此基础上,观察上述模型大鼠MI激活情况及其激活所释放的细胞因子和氧化应激产物、神经递质的改变,并通过细胞培养实验探讨应用抗炎药物米诺环素和抗精神病药物后的治疗效果是通过上述哪些通路实现的。本研究将首次论证MI在精神分裂症阴性症状中作用的可能信号通路,为精神分裂症阴性症状的病因学研究及新的治疗靶点提供基础实验证据。

英文摘要: The pathological mechanism of negative symptoms of schizophrenia is unknown. There is no effective treatment for negative symptoms and some patients had severe brain functional disability. Recently, some research consider that the neuroinflammation induced by over-activated microglia may be one of the pathogenesis of negative symptoms. NF-κB, MAPK and JAK-STAT signaling pathways are related with neuroinflammation. We established an microglial activation animal model on schizophrenia in our pilot study. On this basis, we are supposed to examine the microglia, changes of cytokins, oxidative stress products and neurotransmitters in that model; on the other hand, we examine which pathways mentioned above involved in the effect on negative symptoms of minocycline and antipsychotic drugs. This is the first time to verify the possible signaling pathways that microglia involved in negative symptoms of schizophrenia, and to provide evidences for etiology and new therapeutic target of schizophrenia.
