中文摘要: 牛结核是由牛分支杆菌和结核分支杆菌等结核分支杆菌复合群引起的一种人兽共患的慢性消耗性传染疾病,严重威胁人畜健康。Ipr1基因已经被证明能介导巨噬细胞以细胞凋亡的方式限制结核分支杆菌增殖,但其作用机制尚未被阐明。本研究拟结合转录组学与分子生物学技术研究Ipr1介导的巨噬细胞凋亡途径,以期阐明Ipr1抗结核分支杆菌的作用机理。首先进行表达谱和small RNA测序,筛选参与细胞凋亡信号通路的差异表达基因和microRNA作为候选基因,随后利用western blot, qPCR, RNAi和共免疫沉淀(COIP)技术研究候选基因和microRNA的功能及其与Ipr1的调控关系,借以绘制Ipr1参与的巨噬细胞凋亡转录调控网络。最后研究转Ipr1基因牛巨噬细胞对牛分支杆菌的抗性,验证绘制的信号调控网络。本研究将为结核病的预防和抗病转基因家畜育种研究提供依据和新思路。

英文摘要: Bovine tuberculosis (BTB) is a chronic animal-human infectious disease caused by the pathogen Mycobacterium bovis and poses a long-standing threat to human and livestock worldwide.The fact that Ipr1 limited proliferation of Mycobacterium bovis via switching necrotic to apoptotic death pathway has been proved in macrophages, however, the mechanism is unkown. This research is intended to understand the molecular mechanism of Ipr1 resisting Mycobacterium bovis by combining the genomic and moleculai biological technologies. Digital Gene Expression profiling and small RNA sequencing will be employed to screen the potential candidate gene and microRNA invovled in apopotosis, then verify the functions and uncover the relationship between Ipr1 and the candidate gene and microRNA, an Ipr1 mediated regulatory network will be generated. Finally, we will investigate the macrophages resistance of Ipr1 transgenic cows to validate the regulatory network. This research will provide new insights and solutions for tuberculosis therapy and disease-resistant transgenic animal studies.
