中文摘要: 动脉粥样硬化是动脉内皮损伤后的慢性炎症性疾病。转录因子参与动脉粥样硬化的形成和发展。新发现的转录因子Tbx20主要表达在心血管系统,参与胚胎期心血管发育和成年后心血管功能维持,但它在血管内皮细胞损伤乃至动脉粥样硬化发生中的作用目前尚未见报道。我们前期的研究发现:人脐静脉内皮细胞在ox-LDL刺激下,Tbx20表达下降,细胞凋亡增加;而过表达Tbx20后PPARγ表达升高,细胞的凋亡显著下降。由此我们提出假说:Tbx20可能通过调控PPARγ表达保护内皮细胞,抑制动脉粥样硬化的发生和发展。本研究拟从分子、细胞和整体水平,利用人脐静脉内皮细胞及动脉粥样硬化动物模型,以基因过表达、RNA干扰、免疫组化、ChIP、生物信息学分析等方法,明确Tbx20在血管内皮细胞损伤中的保护作用,探讨其作用机制。本研究将从Tbx20这个新视点为揭示动脉粥样硬化的发生机制奠定基础,为防治动脉粥样硬化提供新思路。

英文摘要: Atherosclerosis is a chronic inflammation disease which is initiated by the endothelial cell injury. Many transcription factors take part in the initiation and progression of atherosclerosis. As a transcription factor mainly expressed in cardiovascular system, Tbx20 plays an important role in embryonic cardiovascular system development and adult cardiovascular biological function maintenance. However it is still not clear about the role of Tbx20 in endothelial cell injury and atherosclerosis. Our previous study found that ox-LDL could induce Human Umbilical Vein Endothelial Cell (HUVEC) injury and apoptosis with decreased Tbx20 expression. Overexpression of Tbx20 by adenovirus vector could increase PPARγ expression and decrease HUVEC apoptosis. Thus, we hypothesize that Tbx20 could decrease endothelial cell injury and attenuate atherosclerosis development by regulating PPARγ expression. We will use adenovirus vector gene transfer, RNA interference, immunohistochemisry, Chromatin Immunoprecipitation and bio-informatics methods to investigate Tbx20's role and transcription regulation mechanism in molecular, cellular, tissular and animal levels during the development of atherosclerosis.
