中文摘要: 本项目主要对两种湖北麦冬均一多糖单体LSP1和LSP2的降脂疗效及其作用机制进行研究。从整体上探讨LSP1和LSP2对脂质代谢的影响。体外3T3-L1细胞实验使用蛋白试剂盒,检测LSP1和LSP2对脂肪细胞中游离脂肪酸FFA等脂质代谢相关因子的活性。体内实验中,将高血脂动物(SD、Zucker大鼠)随机分为高血脂对照组、阳性组、正常组、LSP1、LSP2(高、低剂量组)。28天后测定各组给药前后的血清胆固醇相关指标。取各组动物肝、肾、中动脉血管组织等,病理染色,以观察其病理形态。使用免疫组织化学、蛋白印迹电泳法Western Blot和Realtime-PCR 等方法检测湖北麦冬均一多糖对PPARγ/CD36/oxLDL/ABCA1/ABCAG以及PPARγ/AFABP/ACS/FATP/LPL/ApoA,B,C,E两条信号通路的蛋白表达的影响,从分子水平上探讨其改变机体胆固醇代谢的机制。

英文摘要: This study was to investigate anti-hyperlipidemic effect and its mechanism of polysaccharides from the tuberous root of Liriope spicata (Thunb.) var. prolifera activity of LSP1 and LSP2(LSP1:Liriope spicata polysaccharides 1 and LSP2:Liriope spicata polysaccharides 2)on the Zucker rat, 3T3-L1 adipocyte. We also evaluate the effect of TLSP, LSP1 and LSP2 on hyperlipidemic patient. The level of FFA, Leptinof adipocytes are estimated by kit , respectively. The Zucker rat are received daily oral administration of the LSP1 and LSP2 for 28 days after hyperlipidemia animal model is established. TC, TG, HDL, LDL and VLDL are performed to evaluate the anti-hyperlipidemic activity of polysaccharides. In addition, Oil-red O staining analysis of liver, kidney,white and brown adipose are performed to observe the pathological process of each group. To evaluate the mechanism of lipid-lowering activity of the polysaccharides , the proteinand and mRNA expressions of PPARγ/CD36/oxLDL/ABCA1/ABCAG 以及PPARγ/AFABP/ACS/FATP/LPL/ApoA,B,C,E in liver tissue or mcrophagesare of animal model are estmited by the immunohistochemistry,immunofluorescence, western blotting and Realtime-PCR after 28 days of LSP1- and LSP2-administration.
