中文摘要: 沙棘为典型的多基原藏药,除中国药典收载的Hippophae rhamnoides L.外,西藏沙棘H.tibetana Schlecht.等8种沙棘属植物及其变种均作为藏药使用,严重影响药材的质量控制及临床使用的疗效等同性,传统生药学方法对多基原沙棘质量控制十分困难。本项目在前期国家自然科学基金(81303310)等工作基础上,结合藏医药理论和用药经验,提出EST-SSR和整体代谢谱分析,能实现多基原藏药沙棘品种鉴定及整体性质量控制的假说。筛选新型EST-SSR分子标记,对多基原沙棘进行分子鉴定。整合运用1H-NMR、UPLC-Q-TOF/MS和GC-MS等代谢谱分析技术,系统表征沙棘的整体性代谢产物。以止咳、祛痰、血小板聚集等试验,对多基原沙棘进行药效学评价。采用典型相关分析处理,揭示多基原沙棘品种的遗传型、化学型和药效型之间的相关性,为藏药沙棘鉴定和质量控制提供科学依据。

英文摘要: Seabuckthorn is a typical Tibetan medicine with multiple botanical origins, besides Hippophae rhamnoides L. which contains in Chinese Pharmacopoeia, there are eight seabuckthorn genus and its variants are used as Tibetan medicine, seriously affected its quality control and effect of equality in clinic. There is serious problem to identify them with conventional pharmacognostical methods. This project proposed hypothesis EST-SSR and overall metabolic profiling can identify and control the entire quality of Tibetan medicine seabuckthorn based on the early National Natural Science Foundation of China (81303310) and combined with the theory and experience of Tibetan medicine. The project use RNA-seq to obtain EST sequences, screening new EST-SSR markers to identify seabuckthorn genus and its variants. Integrating UPLC-Q-TOF/MS,1H-NMR and GC-MS metabolic methods to characterize its metabolites systematically, using cough-relieving and sputum-removing trials in mice and platelet aggregation trials in rats to evaluate the pharmacodynamics of seabuckthorn. Investigating the correlation between genetic information, chemical composition and pharmacodynamic efficiency of different origins of seabuckthorn by canonical correlation analysis method, providing scientific basis for identifying and controlling the quality of Tibetan medicine with multiple botanical origins-seabuckthorn.
