中文摘要: 嗅鞘细胞(OECs)促进中枢神经系统神经功能修复作用突出,与其本身所具有的特性有关,但来源非常有限,自体移植需要开颅取材,损伤大风险高,而异体移植存在伦理及免疫排斥问题。雪旺细胞(SCs)取材方便,是进行自体移植的优秀种子细胞,但其促进中枢神经系统神经功能修复作用较弱,本研究试图在成熟体细胞诱导转化方面进行研究,探索由SCs转化来的OECs(ST-OECs)细胞培养节点及培养基配方,观察ST-OECs在体内外脊髓实质内存活、迁移、再髓鞘化、穿越胶质瘢痕的特点,同时探讨其促进神经再生作用机制,为自体ST-OECs细胞移植治疗晚期脊髓损伤解决细胞来源问题并兼顾治疗效果。

英文摘要: The remarkably function of neuroregeneration of olfactory ensheathing cells (OECs) bear on its special biological property. However the source of OECs is very limited. There are need craniotomy to obtain the OECs for autologous cell transplantation, so it has very high risk for patients of spinal cord injury, and the ethical problems also is the obstacle for allografts cell transplantation as well as the immunologic rejection . Schwann cells (SCs) conveniently, is a important seed cells for autologous cell transplantation to the patient which have peripheral nerve injur, but its function of neuroregeneration in the central nervous system is very weak. We attempts to induced the transformation of SCs to OECs(ST-OECS), and observe the survival, migration, neuroregeneration, remyelination, et al. which transplantated the ST-OECs into the spinal cord parenchyma in vivo and in vitro. To investigate the mechanism of promote nerve regeneration and to provide experimental data for clinical application.
