中文摘要: 生殖干细胞是人类和动物生殖系统的源头,在种族延续、配子发生等生命过程中起决定性作用,生殖干细胞发育的研究一直是当前发育生物学领域中的核心问题之一。生殖干细胞的发育受到严格地调控,翻译后水平的调控是其中一个关键的环节。蛋白质泛素化是翻译后调控的一种重要形式,蛋白质泛素化对于生殖干细胞发育调控的分子机制我们目前所知甚少。本项目将以果蝇雌性生殖干细胞为模型系统,在已有的工作基础上,运用遗传学、分子生物学和细胞生物学等手段,研究E3连接酶Gcn5在果蝇雌性生殖干细胞命运决定中的作用机制。我们希望通过本项目的开展,加深我们对生殖干细胞发育的基础理论的理解,同时也希望本研究为生殖生物学临床基础研究和生殖疾病相关的新药开发提供指导。

英文摘要: Germline stem cells are the origins of reproduction system in human and animals and essential for ethnic continuity and gametogenesis.Studies on germline stem cells(GSCs) are the focus in the field of developmental biology.The development of germline stem cells is regulated accurately and the post- translational regulatory is very important. Protein ubiquitination is one of the post-translational modifications and much less is known about the regulatory mechnism of ubiquitination in the development of GSCs.In this study, we'll focus on the project: the regulatory mechanism of Gcn5, a E3 ligase, in the GSC maintenance on the model of Drosophila ovary by the method of genetics, molecular biology and celluar biology.We expect that from this project we'll take a deep understand in the development of GSCs and our results could provide guidance on the clinic and the research and development of new medicines related to the reproduction system diseases.
