中文摘要: 天然产物开发在抗肿瘤药物的研究中占据十分重要的位置。极地真菌长期处于极端生存环境并进化出独特的代谢系统,具有生成新奇化学骨架和活性多样的次级代谢物之潜力。对极地真菌次级代谢物的化学和抗肿瘤活性研究必将为抗肿瘤药物的研究注入全新活力。本课题组与中国极地研究中心密切合作,经对后者提供的15株来自北极的珍稀真菌抗肿瘤生物学活性筛选,首次选取三株活性较好、次级代谢产物丰富的真菌菌株D-1、D-2、D-3为研究对象,综合运用天然药物化学、微生物学、分子药理学等学科专业技术,对其抗肿瘤活性次生代谢产物进行系统的化学结构和抗肿瘤生物活性研究,并将选取活性最好的化合物进行初步抗肿瘤信号通路和作用机制研究,以源头创新促进我国对极地微生物资源的利用和具有自主知识产权抗肿瘤新药开发。此外,极地微生物次级代谢产物的研究对于丰富天然产物库也具有现实意义。

英文摘要: It's well known that the development of natural compounds show a very important role in studying the antitumor drugs. Polar fungi grow in the extreme environment and possess the characteristic metabolite systems, which will produce the unique chemical skeletons and diversity biological metabolites. The study of the secondary metabolites and the antitumor activities of polar fungi will inject new vitality for the research of antitumor drugs. Our group established the close cooperation with Polar Research Institute of China. On the basis of the screening antitumor activites of 15 fungi derived from Arctic region, 3 fungi (D-1、D-2、D-3) with good biological activites and abundent secondary metabolites were picked out. This project will focus on the study of the antitumor secondary metabolites of 3 strains and their antitumor activities by integrating of natural medicinal chemistry, microbiology, molecular pharmacology professional technology, and the preliminary study of antitumor signal pathway and biological mechanism of the good biological metabolites.
