中文摘要: LincRNA是一类起重要功能且与疾病密切相关的分子。申请人发现lncRNAs中的PANDA基因在职业三氧化二砷暴露人群中表达显著增高,且与二甲基化指数显著相关。生物信息学软件预测PANDA启动区存在多个可能的甲基化区域,在结合文献的基础上提出:PANDA启动区甲基化区域,将影响基因表达,砷通过这两种机制影响PANDA表达。本研究首先检测职业三氧化二砷暴露人群和对照人群外周血淋巴细胞中PANDA基因启动区DNA甲基化水平和PANDA的表达量,初步探索三氧化二砷暴露下甲基化改变对PANDA表达影响。然后通过三氧化二砷染毒细胞比较染毒前后启动区甲基化水平和PANDA变化证明砷对二者的影响。最后利用干扰细胞中甲基化变化表达,观察三氧化二砷染毒后细胞中PANDA的变化,证明三氧化二砷通过甲基化影响PANDA的表达。

英文摘要: LncRNA is an important functional molecule associated with many diseases. PANDA gene which belongs to lncRNAs is expressed at very high level in human exposure to arsenic trioxide. Furthermore, PANDA gene is related to secondary methylation index apparently. It is estimated that there were many possible methylation area in promoter of PANDA by the bioimformatics software. methylation area of PANDA may influence gene expression, by which arsenic trioxide improves the PANDA expression. In this project, the promoter DNA methylation level of PANDA and PANDA in the arsenic trioxide exposure population and control will be detectived in order to find if arsenic trioxide affects the expression of PANDA by changing methylation . Through arsenic trioxide exposing cells, changes of promoter DNA methylation level of PANDA and PANDA of cells under arsenic trioxide exposure will be observed to prove the influences to promoter DNA methylation level of PANDA and PANDA by arsenic trioxide. Finally, the expression change of PANDA under arsenic trioxide exposure will be observed to prove that arsenic trioxide influences the expression of PANDA through methylation
