中文摘要: 瘢痕修复是当前亟待解决的医学难题之一,探索瘢痕的形成机理及其治疗方法成为分子生物学、生物物理学、生物医学工程学、组织工程学和临床医药学等学科的研究热点。本研究采用驻极体技术和药剂学技术,研制驻极体、5-氟尿嘧啶和驻极体5-氟尿嘧啶贴剂,从电场与药物粒子的相互作用出发,研究驻极体和驻极体载药系统的电荷储存稳定性,各类载药系统中药物贮存、释放规律、皮肤深层组织中的分布及其它们的影响因素。利用兔耳增生性瘢痕模型,对瘢痕的关键生化指标I型胶原、III型胶原、成纤维细胞、转化生长因子、半胱氨酸天冬氨酸蛋白酶、基质金属蛋白酶组织抑制因子和基质金属蛋白酶等进行研究,首次探讨驻极体及驻极体载药系统对瘢痕组织形态学、分子生物学变化规律的影响,阐明驻极体治疗增生性瘢痕的机制,并为驻极体在生物医药学方面的应用开拓新的领域,为创面治疗和瘢痕修复提供一种新的思路和科学依据。

英文摘要: Scar repairing is one of the difficult medical problems.Exploring on the mechanism of scar formation and therapy has been the hot research topics of molecular biology, biological physics, biomedical engineering, tissue engineering, clinical medicine and pharmacology. In this study, we use electret technology and pharmaceutical technology to prepare the electret, 5-fluorouracil (5-Fu) transdermal system (drug system) and electret 5-Fu transdermal system (electret drug system). The charge storage stability, drug storage and release behaviors of the electret and electret drug system, drug distribution in varies skin tissue, as well as the influence factors are to be studied from the aspect of interaction between electric field and drug molecules. A hypertrophic scar (HS) model in the rabbit ear is to be produced. And some key biological parameters, such as collogens I and III, fibroblasts, transforming growth factor (TGF-B1), cysteine-containing aspartate-specific proteases (caspase)-3, caspase-9, P311, tissue inhibitor of metalloproteinases (TIMP), matrix metalloproteinase (MMP)etc., are to be studied to explore the mechanism of electret on scar repairing. We hope our study can open new electret frontiers in biomedical application, and provide new ideas and scientific evidence for wound healing and scar repairing.
