中文摘要: 动脉粥样硬化是严重危害人类健康的常见病,oxLDL激活单核/巨噬细胞生物学功能对动脉粥样硬化发生发展具有关键意义。迄今对oxLDL激活单核/巨噬细胞功能的关键内在机理仍知之甚少。我们前期研究发现,miR-9在调控oxLDL激活单核/巨噬细胞脂质摄取、炎症因子分泌、细胞迁移及凋亡等过程中起重要作用,表明其对动脉粥样硬化斑块形成发展有重要意义。因此,我们拟在前期研究基础上,通过转基因技术上调/下调miR-9表达,构建单核/巨噬细胞特异性过/低表达miR-9斑马鱼模型,体内进一步研miR-9对单核/巨噬细胞功能及动脉粥样硬化的影响。此外,在已进行体外生物信息学预测并初步验证的基础上,通过RISC-Sequencing技术进一步发现并证实其下游靶基因,以期找到调控oxLDL激活单核/巨噬细胞生物学功能的更关键靶点,为动脉粥样硬化防治提供新方向。

英文摘要: Atherosclerosis is a common disease that is detrimental to human health. Monocyte/macrophage function stimulated by oxLDL is essential for the origin and development of atherosclerosis. However, the mechanism involved is not clear yet. Our earlier vitro research has demonstrated that miR-9 plays a vital role in ahtrosclerosis , which functions by regulating monocyte/macrophage lipid intaking, inflammation factors secretion, migration,as well as cell apoptosis. Thus, we establish a monocyte/macrophage specific miR-9 expression zebrafish model in attempt to carry on a further vivo study. This vivo study is on purpose of verifying the role of miR-9 in monocyte/macrophage function and atheroscleroma formation. Furthermore, on the ground of earlier bioinformatics prediction and provement in vitro, RISC-Sequencing will be further applied to screen for downstream genes of mir-9, which will help to find some more crucial targets in atherosclerotic disease treatment.
