中文摘要: 急性肺损伤是常见呼吸道重症疾病,死亡率很高,现有的治疗方法疗效有限。近几年研究发现骨髓间充质干细胞可以改善肺损伤的严重程度,但其机制不明。新近研究发现,特络细胞(telocytes)能够诱导、促进干细胞的生长、分化,可以作为干细胞的辅助细胞;对特络细胞的基因谱分析发现其高表达Rbp1基因。本课题拟用特络细胞联合骨髓间充质干细胞体内治疗急性肺损伤,通过检测各种相关的细胞表面标记物、信号转导蛋白、趋化因子、细胞因子等的表达,分别探索上述细胞之间的相互作用,阐述Rbp1在特络细胞联合骨髓间充质干细胞体内治疗急性肺损伤中的调节作用,揭示其中的分子机制及信号通路,丰富和深化对特络细胞生物学功能的研究和认识,为深入开展以特络细胞为基础治疗肺损伤奠定实验和理论基础。

英文摘要: Acute lung injury is a common pulmonary severe disease with high mortality, and the current therapies are limited and lack of efficacy. In recent years, more and more studies identified that bone marrow mesenchymal stem cells(MSC)could improve the severity of lung injury, but the mechanism is still unclear. Recent studies confirm that telocytes could induce and improve the growth and differentiation of stem cells, and play a role as stem cell helper cell; study from genomic analysis reveals that telocytes highly express Rbp1 gene. In the present project, we will treat acute lung injury by combining telocytes with MSC in vivo, and in further, we will detect the expressions of special biomarkers on the surface of cells, signal transfactor proteins, chemokines and cytokines, to explore the interactions among these cells, to explain the regulative effects of Rbp1, to reveal the potential mechanism and the signal pathway during this process, to improve the study of cell biological functions of telocytes, and try to provide experimental and theoretical foundation for the further in vivo study in the treatment of lung injury with telocytes.
