中文摘要: 发展直接用于复杂生理样品中重金属离子、小分子等检测的生物传感器是当今科学研究的热点和挑战,基于DNAzyme的生物传感器由于高灵敏、高选择性已被广泛地应用于各种重金属离子、小分子等的检测中。但是,目前极少基于DNAzyme的生物传感器能够直接用于复杂生理样品的检测,而DNAzyme的体内应用性能迄今未有报道。本项目拟基于DNAzyme自组装纳米材料发展直接用于复杂于生理样品中重金属离子、小分子等检测的新型DNAzyme荧光纳米探针。DNAzyme自组装纳米材料策略,具备了DNAzyme的高灵敏、高选择性和纳米材料保护核酸免受样品中核酶降解或干扰的优势,从而能够实现对复杂生理样品中重金属离子、小分子等目标物真正快速、高灵敏、高特异性检测,为上述物质的临床医学检验提供有价值的理论依据和技术支持。同时,用于活细胞中的荧光成像研究,为DNAzyme的体内性能研究提供有价值的理论参考和技术支持。

英文摘要: The development of biosensors for heavy metal ions and small molecules direct detection in the complex physiological samples is the challenges of today's scientific research. Due to the high sensitivity and selectivity, DNAzyme-based biosensors have been widely used in various heavy metal ions and small molecules detection. However, very few DNAzyme-based biosensors can be directly used for the detection of complex physiological samples at present, and the in vivo application performances of DNAzyme has not been reported so far. The project intends to develop the novel DNAzyme fluorescent nanoprobes based on DNAzyme self-assembled nanomaterials,which can be directly used for the detection of targets such as heavy metal ions, small molecules in complex physiological samples. With the high sensitivity and selectivity of DNAzyme, and the ability of nanomaterials protect the nucleic acids from nuclease digestion or interferences, the DNAzyme-assembled nanomaterials strategy can be applied for the really fast detection of heavy metal ions and small molecules in the complex physiological samples with highly sensitivity and specificity, which will provide a valuable theoretical basis and technical support for the detection of the above-mentioned targets in clinical medicine.
